Application | Methyl 5-fluoro-2-hydroxybenzoate is an organic intermediate, which can be obtained by esterification of 5-fluorosalicylic acid and methanol. The esterification method can be sulfuric acid dehydration or 5-fluorosalicylic acid is first made into acid chloride and then acylated. |
preparation | at 0 ℃, slowly add concentrated sulfuric acid (20mL) to the stirring solution of 5-fluorosalicylic acid (25g,160mmol) in MeOH(250mL). The resulting reaction mixture was refluxed for 48h, then concentrated under reduced pressure and the resulting crude was alkalized with saturated NaHCO3 to pH 8.0. The mixture was neutralized with a 1.5N HCl solution and extracted with EtOAc. The organic layer was dried with anhydrous Na2SO4 and concentrated to give methyl 5-fluoro-2-hydroxybenzoate as a light brown liquid (22.8g,83% yield). |
